There's no cardroom that's more convenient to go to than Casino at the Empire. Located smack in the middle of Leicester Square, open 24/7, and you don't even need to become a member to play. There's always action, and very loose action at that.
This cardroom was previously known as The Vic, and is a poker landmark in the UK, and the spot for high stakes cash games in London. It's now been rebranded as The Poker Room. It's extremely well-run, and boasts the widest variety of games and stakes you will find in London. It's not unusual to spot the odd poker pro playing at one of the tables.
Previously the Gala Casino Queensway, this casino offers purely cash games. The location is pretty central, so if you are looking for some low stakes poker and don't feel like travelling too far to play, this place is worth trying.
The Hippodrome Casino, located centrally in Leicester Square, opened in July 2012 and offers a nice mix of cash game action.
The River Card Club is located way up in Stanmore in North London. It's quite a small cardroom, with a very friendly and social atmosphere. If you live in North London, this cardroom is worth checking out.

Aspers Casino in Westfield Stratford City is the largest 'supercasino' in Britain, and the 15-table poker has a solid tournament schedule and lots of soft cash action.
It's not strictly in London but it is close enough for us to mention here on Londonpokerguide. This is a decent private members club with good comps and some interesting promotions.

Cheap stakes and central location - that's pretty much what you get at the G Casino Piccadilly
Centrally located, catering to higher stakes players. No tournaments being held at the moment.
